Melanotan-1 (afamelanotide) is a synthetic version of alpha-MSH that mainly activates the MC1 receptor, which controls melanin production in pigment cells. Researchers use it to study pigmentation and how melanin production is regulated.

About This Compound

About Melanotan-1

Melanotan-1, known by the international nonproprietary name afamelanotide, is a synthetic analogue of alpha-melanocyte-stimulating hormone. It keeps the full 13-residue length of the natural hormone and changes exactly two positions: methionine at position 4 becomes norleucine, and the L-phenylalanine at position 7 becomes its D-enantiomer. Both substitutions target the same weakness - native alpha-MSH is taken apart by peptidases within minutes - and together they give a molecule that survives long enough to be useful as a laboratory tool. It is worth reading alongside Melanotan-2, which solves the same stability problem a different way: MT-2 is a shortened CYCLIC peptide, where MT-1 stays linear and full-length. Two Substitutions

Nle4 And D-Phe7

Native alpha-MSH is degraded rapidly by peptidases. Replacing Met4 with norleucine removes the oxidation-prone sulfur, and inverting Phe7 to the D-form puts a bond that proteases are not shaped to cut into the middle of the recognition sequence. Published work reports the pair together produce a markedly longer-lived and more potent analogue than the parent hormone.

Where The Selectivity Differs
